Guggulsterones function as promiscuous nuclear receptor modulators: FXR antagonism increases hepatic LDL receptor expression, PXR activation enhances drug/toxin metabolism, and thyroid-stimulating activity may enhance T4-to-T3 conversion via hepatic deiodinase upregulation. Anti-inflammatory effects occur through inhibition of NF-κB nuclear translocation and STAT3 phosphorylation. Ayurvedic Philosophy

Guggulu (Commiphora mukul) is the only Dravya (substance) in Ayurveda that lends its name to an entire class of formulations — the Guggulu Kalpas. The Sushruta Samhita describes Guggulu's Shodhana (purification) in extensive detail, emphasizing that raw Guggulu must be processed with specific herbal decoctions (Triphala Kwatha, Vasaka Kwatha, or milk) to remove toxic ketosteroids while preserving therapeutic guggulsterones. Classified simultaneously as Lekhana (scraping), Medohara (fat-reducing), Vatahara (Vata-pacifying), and Rasayana (rejuvenative), Guggulu occupies a unique position in Dravyaguna Vijnana — it is one of the few heating, pungent substances that is also considered a Rasayana. The Yogaratnakara declares that no Vata disorder is beyond Guggulu's therapeutic reach.
